Pratical 7 (week 7)

Previously, all MBB students have done the transformation procedure. To someone who don't know what we r doing, basically MBB Sunway students are trying to clone amylase gene from Bacillus subtilis. Now, we have done the transformation part and hence we need to do some testing to make sure the transformation gene has been ligated with the vector and the vector itself must be transformed with the E. coli that act as host. Since transformation occured spontaneously, not all students manage to have transformation bacteria. From 240 students, only 6 clones manage to be transformed.

To check the transformation present or not, a simple test by using iodine was done. THe bacteria was growth on Nutrient agar containing starch and after a few days, the colonies growth on the plate. Then, iodine solution was poured on the plate. oppss, before doing this, some sample are picked from the agar lah... sbb nanti mner nak continue experiment, lps mskkan iodin matila bacteria2 itu.

ok, pas amik samples n transfer kat another plates, iodine test is done to make sure there is transformation bacteria or not. Luckily in my group, ade la mununjukkan positive result on iodine test. camner positive ditentukan??? kan aku dah kate agar tu ade starch, so klu bacteria tu ade amylase gene (transformation bacteria), maka bley la die merembeskan enzim amylase untuk menghazam kanji. maka, dapat kita lihat pada plate yang ada koloni itu semacam halozone....

ok, since ak nyer positive, spttnya koloni2 yang ak dah simpankan td akan membesar dan sepatutnya mempunyai transformant bacteria.... tp malangnya ntah knapa xde. ok, ini baru rapuan tentang transformation jer.... ak dah malas... babai, mase ntuk wat repot tu:P
